Ajax functionality is integrated in ASP.NET 3.5 SP1 and does not require any additional downloads.

To use Ajax, login to DNP (your control panel aka. DotNetPanel) -> Web Sites section and click on the website you want to use it with. Then click on the Extensions tab, and change the ASP.NET from the drop down to "2.0 Integrated Pipeline" (aka. 3.5).

That will allow you to use Ajax without any additional assemblies.

Published a 3.5 ASP.Net web application and am getting errors that AJAX is not available. I am using 2.0 Integrated Pipeline. This did not work. Please help. I need to get a site published ASAP.

Please note that AJAX binaries are by default installed across all of our servers. Therefore, it is an issue with the application or the settings within your web.config. I suggest opening up a support ticket so we can further look into this for you.
